2,4,5-Trichloropyrimidine2,4,5-Trichloropyrimidine is used in the synthesis of potent and selective anaplastic lymphoma kinase (ALK-5) inhibitors, used as an anti-tumor treatment. It is also utilized in the manufacture of EGFR inhibitors. A route to 4,5,6-trichloropyrimidine-2-carbonitrile was developed starting from 4,6-dichloro-2-(methylthio)pyrimidine. The latter was converted to 4,6-bis(benzyloxy)-5-chloropyrimidine-2-carbonitrile in four steps giving an overall yield of 67%. The process involved nucleophilic displacements of the chlorine atoms in benzyloxide and then the oxidation process of the sulfide sulfide to sulfone, and its displacement through the cyanide group and the chlorination of the pyrimidine C5 location using NCS. 4,6-Bis(benzyloxy)-5-chloropyrimidine-2-carbonitrile was finally converted into 4,5,6-trichloropyrimidine-2-carbonitrile in a moderate (30%) yield in a two-step procedure.
